Stored Stallion Sperm Quality Depends on Sperm Preparation Method in INRA82 or INRA96
نویسندگان
چکیده
Removal of seminal plasma facilitates stallion sperm survival during storage, but washing may damage chromatin. Therefore, quality was compared in samples following single-layer centrifugation (SLC) or and controls (extension only) two extenders, INRA82 INRA96. Ejaculates from six stallions were split among treatments: SLC, washing, controls, Sperm motility acrosome status evaluated at 0, 24, 48, 72 96 hours; morphology hours chromatin integrity 0 hours, with storage 6°C. INRA96 had better motility, status, normal than INRA82. The SLC higher fewer reacted acrosomes lower fragmented washed samples. Fewer spermatozoa tail defects observed after washing; those In conclusion, (except for morphology) controls. method is a useful adjunct to preparation, especially before artificial insemination.

Effect of method and clinician on stallion sperm morphology evaluation.
The objective of this study was to determine the effects of method and clinician on stallion sperm morphology evaluation. Five clinicians evaluated 60 semen samples using wet-mount preparations with phase-contrast, eosin/nigrosin-stained semen smears, and Papanicolaou-stained semen smears. There were significant differences among methods for all sperm morphology categories and most intra-class ...
متن کاملMeasurement of Sperm Concentration in Stallion Ejaculates Using Photometric or Direct Sperm Enumeration Techniques
A highly significant correlation was detected among photometric and direct sperm enumeration techniques for evaluating sperm concentration. Depending on the instrument used, the photometric techniques tended to overestimate or underestimate sperm concentration when compared to a hemacytometer standard. Precautions should be taken when using techniques other than a hemacytometer to measure sperm...
متن کاملStallion Sperm Selection by Density Gradient CentrifugationInvolving a Double Layer Colloid: Effects on Sperm Subpopulation Dynamics in Fresh and Stored Semen
Sperm selection techniques have became an important tool to improve sub fertile ejaculates. The main aim of this study was to assess the dynamics of motile sperm subpopulations of ejaculates from subfertile stallions subjected to density gradient centrifugation (DGC) involving a double layer colloid [DGC2], before and after storage for 24 and 48 hours. Ejaculates from eight stallions with ferti...
متن کاملSperm Function Tests in the Stallion
Introduction: There are many tests that describe semen quality in the stallion which include sperm motility, morphology, and longevity of motility and determination of total sperm numbers. In addition, recently, the flow cytometer (FC) has been used to evaluate various sperm compartment following staining with specific fluorescent probes that bind to different compartments of the sperm. Our lab...
متن کاملCooling of ejaculated and epididymal stallion sperm
After a serious injury or sudden death, epididymis cauda sperm recovery and cryopreservation may present as the last opportunity to obtain genetic material from a valuable stallion. This study evaluated the viability of cooled equine sperm collected by three different methods: sperm of ejaculated (G1), sperm recovered from the epididymal cauda immediately after orchiectomy (G2) and sperm recove...
